I am setting up a new domain(s) and would like to use an existing Office 365 Home Premium subscription.
Is this possible? Or do I need to have a Business subscription to 365?

Based on looking at https://products.office.com/en-us/compare-all-microsoft-office-products?&activetab=tab:primaryr1
The Home Editions do not include email hosting - they do include outlook, but not email at the domain, you would have to upgrade to the business edition to get the email box as well.
I would contact Microsoft Support (or wherever you bought the license through) to see if it is possible to just upgrade your account or if you need to cancel it and purchase a new account
